Application summary

・目標近年、Cardio-Oncology分野に注目が集まっており、薬剤心毒性の予測・評価は重要な課題として認識されている。一方で、薬剤性心毒性は、不整脈に加えて、収縮毒性、構造的な毒性など多岐にわたる。また、心筋細胞活動電位には種差があることが知られており、動物試験のヒトへの外挿性は限られていると考えられている。これらの要因から、薬剤性心毒性の新たな評価法の開発が求められている。本邦にて発見されたiPS細胞から作成されたiPS心筋を用いた評価法は、抗がん剤による心臓への毒性評価を含めて多岐にわたる薬剤性心毒性の予測や種差の問題を克服できる可能性があるため、本研究において評価法の更なる改善、拡大を目指す。また、現在までのICH S7Bにおける議論を踏まえて、当該評価法が国際標準化されるよう働きかけ、本邦発の技術で薬剤性心毒性の評価における国際調和を推し進めることで、国際的な医薬品開発に貢献する。・求められる成果 ①iPS心筋を用いて新たな薬剤性心毒性評価法を開発する。 ②iPS心筋を用いた評価法が国際的に活用されるよう、国際的な標準化を目指す。
